ABSTRACT:   Mottled amago salmon Oncorhynchus masou ishikawe with both yellow- and dark-pigmented skin occurred together with typical albino individuals in a commercial farm. Out of 12 mottled fish examined by DNA content flow-cytometry and erythrocytic nucleus size, three were diploid, eight were haploid-diploid mosaic and one was diploid-triploid mosaic. This fact indicates that the mottled coloration might link to polyploid mosaicism. Genotype of diploid and non-diploid cells at the albino locus was estimated in nine mature mottled fish by observing the frequency of wild-type and albino progeny when mating to homozygous albino ( aa ). One diploid and three haploid-diploid mosaic mottled fish were presumed to have mosaic genotype with both hemizygous ' a ' and heterozygous ' Aa ' cells ( a/Aa ), because the segregation ratio between two phenotypes was 1 : 1. Three other haploid-diploid mosaic fish were presumed to have mosaic genotype with both hemizygous ' a ' and homozygous ' AA ' cells ( a/AA ), because of exclusive occurrence of wild-type phenotype in the progeny. The diploid-triploid mosaic mottled fish was presumed to have mosaic genotypes ' aa / AAA ', ' aa/AAa ' or ' aa / Aaa ', because this fish yielded only albino progeny. One diploid mottled fish produced both two phenotypes but albino embryos appeared with much more frequency than the expectation, when assuming the genotype ' Aa '. Thus, this fish was considered to have mosaic genotype ' Aa/aa '.  相似文献

ABSTRACT:   The cysts of toxic dinoflagellate Alexandrium tamarense are the seed population for the bloom responsible for paralytic shellfish poisoning (PSP). However, it is impossible to identify the Alexandrium spp. cyst on the basis of morphological features. In this study, we prepared A. tamarense cysts by sexual conjugation in laboratory conditions and developed an efficient DNA extraction method for polymerase chain reaction (PCR) assay. Using the A. tamarense cysts, we established the identification and quantification method showing the species specificity and the high sensistivity for A. tamarense cysts using real-time PCR. This assay was also able to detect and quantify the A. tamarense cysts accurately when mixed with excess cysts of A. catenella (Whedon and Kofoid) Balech prepared by conjugation experiment.  相似文献

ABSTRACT:   The feeding behavior and growth of post-larval Haliotis diversicolor with initial shell lengths (SL) of approximately 500 μm (Exp. 1-1 and 1-2), 800 μm (Exp. 2), and 1200 μm (Exp. 3) were studied in a laboratory setting while they fed on four species of benthic diatom Achnanthes longipes , Cocconeis sublittoralis , Cylindrotheca closterium , and Navicula ramosissima . Exp. 1-1 and 1-2 revealed no marked differences in post-larval growth rates (mean 24–39 μm SL/day) among the diatom species. However, marked differences in growth rates among the species were revealed in Exp. 2 and 3. Three species, A. longipes , Co. sublittoralis, and Cy. closterium , produced faster growth (Exp. 2 mean 29–51 μm/day, Exp. 3 mean 36–44 μm/day) than N. ramosissima (Exp. 2 mean 18 μm/day, Exp. 3 mean 23 μm/day). Post-larvae fed N. ramosissima had lower digestion efficiency (42.8%) than those fed other diatom species (90.7–100%). Diatom extracellular substances appeared to be principally used from post-settlement to 800 μm SL, and diatom cell contents were required to produce rapid growth of larger post-larvae (>800 μm SL). It is likely that the availability of each diatom for post-larvae was affected by diatom morphology, attachment strength, frustule strength, and post-larval size.  相似文献

Although a large number of studies have shown that rats have a serious effect on island ecosystems, questions remain regarding how their invasion alters native species diversity and assemblage patterns on islands. In particular, a long history of invasions makes it difficult to understand the impact of rat invasion. In the present study, I investigate how an increase in predation by black rats (Rattus rattus) alters the assemblage characteristics of land snails on the Ogasawara Islands in the West Pacific. Radical declines in many land snail species have occurred since 2006 due to increasing predation by black rats. The frequency of shells with predation scars was greater on larger species and on those living on the trees and in a shallower position in the litter, suggesting greater predation pressure on these species. As a result of this selective predation, large species living on trees and in shallower parts of the litter declined. Accordingly, when black rats increased on the island, small species and species with a burrowing microhabitat became dominant in the land snail assemblage. The present findings can be used to aid in predicting the effects of rat eradication and in estimating the assemblage patterns and diversity prior to rat invasion.  相似文献

We have constructed a soybean (Glycine max (L.) Merrill) bacterial artificial chromosome (BAC) library from green leaf protoplasts of the cultivar, Misuzudaizu. The library contains 53,760 clones with an average insert size of 116 kb. About 2.9% chloroplast DNA origin was revealed by PCR and colony hybridization. Apart from 2.8% clones having no insert, this library represents 5.2 genome equivalents. With this genome coverage, the probability of having any DNA sequence represented in the library is higher than 99.5%. Three-dimensional pools of the BAC library in combination with the use of a high efficiency genome scanning (HEGS) electrophoresis facilitate rapid and efficient PCR-based screening. An average of five positive clones were identified after screening the BAC library with SSR and STS markers. BAC-end walking was performed for three SSR associated BACs. This library will provide a good resource for positional cloning of agronomically and biologically important QTL genes that Misuzudaizu possesses.  相似文献

OBJECTIVES: To evaluate the ratio of energy intake to basal metabolic rate (EI/BMR) among young female Japanese adults, and to compare the lifestyle and dietary characteristics between relatively low and high reporters. DESIGN: Dietary intakes were assessed over a 1-month period with a validated, self-administered, diet history questionnaire, and lifestyle variables were assessed by a second questionnaire designed for this survey. The ratio of EI/BMR was calculated from reported energy intake and estimated basal metabolic rate. SUBJECTS: In total, 1889 female Japanese university students aged 18-20 years who were enrolled in dietetics courses. RESULTS: Ninety-five per cent of the subjects were classified into a non-obese group (body mass index (BMI) <25 kg m(-2); mean+/-standard deviation (SD): 20.8+/-2.6 kg m(-2)). EI/BMR was 1.43+/-0.40 (mean+/-SD). Sixty-eight per cent of the subjects showed an EI/BMR level below the possibly balanced value of 1.56, 37% showed EI/BMR below the minimum survival value of 1.27 and 2% of the subjects showed EI/BMR exceeding the maximum value for a sustainable lifestyle of 2.4. BMI, body weight and BMR decreased significantly with the increase in EI/BMR (P<0.001). The percentage of energy from carbohydrate was significantly higher, whereas those from fat and protein were significantly lower, among the lower EI/BMR groups. As for food groups, a significantly declining trend from the lowest to the highest EI/BMR groups was observed for cereals. CONCLUSION: Underreporting, rather than overreporting, of energy intake was predominant in this relatively lean Japanese female population. BMI was the most important factor affecting the reporting accuracy of energy intake.  相似文献

Wood ash is a residual material produced during biomass burning. In the northeastern United States up to 80 % of the ash is spread on agricultural lands as a liming amendment with the remainder being disposed of in landfills. As well as raising soil pH, wood ash also adds plant nutrients to soil. This study is an examination of the plant availability of the P in 8 different soils amended with one wood ash. Plant availability was assessed by measuring the biomass and P concentration of corn (Zea mays) L.) plants grown in the greenhouse for 28 d in soil amended with either CaCO3 (control), wood ash to supply 200 mg kg?1 total P, or monocalcium phosphate (MCP) to supply 200 mg kg?1 total P and CaCO3. Both corn growth and P uptake were highest in the MCP treatments, intermediate in the wood ash treatments, and lowest in the controls for all soil types. The soil property which seemed to have the greatest influence on P availability was pH buffer capacity. The soils with the greatest capacity to buffer OH additions also tended to exhibit the greatest absolute P uptake from wood ash-amended soils and the greatest P uptake relative to that from MCP-amended soils. The ability of soil test extractants to predict uptake of P in the three soil treatments was examined. A buffered ammonium acetate extradant overestimated P availability in the ash-amended soils relative to the MCP-amended soils. An unbuffered, acid, fluoride-containing extract provided a measure of P levels that was consistent with P uptake from all soil treatments. The Influence of acid deposition on stream and lake water chemistry was studied in a forested watershed of Hyogo prefecture, Japan. Monthly sampling of four streams, one artificial lake, and precipitation was carried out from 1995 to 2000. The pH of the monthly bulk precipitation and rainwater were ranged from 4.06 to 7.10. No trends were evident during the monitoring periods. The pH and alkalinity in the four streams, which flow into the artificial lake, ranged from 6.37 to 8.72 and 0.077 meqL?1 to 0.485 meqL?1, respectively. The differences in the water quality of the four streams were related to the geology of each watershed. Lower pH and alkalinity were observed during high- discharge periods. On the other hand, the pH and alkalinity of the outflow from the lake ranged from 6.47 to 7.36 and 0.195 meqL?1 to 0.339 meqL?1, respectively. No acidification of the aquatic environment was observed during the investigated periods. The results suggest that this forested ecosystem has the capacity to neutralize incoming acid deposition.  相似文献

Phthalate esters have become widespread contaminants in the aquatic environment, because of their extensive use as non-reactive plasticizers. There is, however, little accurate data on their solubility, transportation, and distribution in the aquatic environment. In this work, we have investigated the influence of humic acid on the water solubility of di-(2-ethylhexyl)phthalate (DEHP), one of the most frequently used phthalate esters in the laboratory studies for DEHP. We have also studied the solid–water distribution of DEHP in the presence of humic acid and particulate matter (activated carbon, ferrihydrite, and kaolinite) to simulate their distribution in a natural aquifer (ternary system). The results show that the water solubility of DEHP can be significantly increased by humic acid. The shape K eq value, the binding constant of DEHP between water and humic acid at equilibrium, was obtained by fitting experimental data for each humic acid. The shape K eq values in the ternary system apparently decreased in the order of ferrihydrite ≤ kaolinite ≈ octanol/water partition activated carbon systems. This result shows that the increase in the hydrophobicity of HA remaining in the solution will lead to the apparent increase of shape K eq in the system since more hydrophilic solid sorbs relatively more hydrophilic HA molecule. The solid–water partition coefficient (shape K W-P ) for DEHP in the environment estimated from this study is consistent with those reported based on the experiments for natural samples. Quantitative values obtained in this study, such as K'ow, shape K eq, and shape K W-P , can be useful for estimating the behavior of DEHP.  相似文献
